ABSTRACT

Objective: To explore the effects of ranitidine on pharmacokinetics of rhein in rats after oral administration of Dachengqi Decoction (DCQD), a compound traditional Chinese herbal medicine. Methods: Twelve male Sprague-Dawley rats were divided into DCQD group and DCQD plus ranitidine group, and were orally administered with DCQD at a dose of 10 g/kg or DCQD (10 g/kg) combined with ranitidine (150 mg/kg), respectively. Blood samples were gathered after a series of time intervals. Metabolism of rhein was determined with a reversed-phase high-performance liquid chromatography with internal standard of 1, 8-dihydroxyanthraquinone and the data were analyzed with DAS 2.1 program. The pharmacokinetic parameters were compared between the two groups. Results: The pharmacokinetic parameters of rhein in the DCQD group, including peak concentration (C(max)), area under the plasma concentration-time curve (AUC), distribution phase half-life (t(1/2alpha)), elimination rate constant (K(10)) and central to peripheral transfer rate constant (K(12)), were significantly different to those in the DCQD plus ranitidine group (P<0.05, P<0.01). There were no significant differences in the other parameters between the two groups. Conclusion: Ranitidine can influence the pharmacokinetics of rhein in rats after oral administration of DCQD.

ABSTRACT

OBJECTIVE: To explore the effects of Tanreqing injection, a traditional Chinese herbal preparation for clearing heat and resolving phlegm, in treatment of acute exacerbation of chronic obstructive pulmonary disease (AECOPD) by improving airway inflammation and airway mucus hypersecretion. METHODS: A randomized controlled trial (RCT) was designed. Ninety AECOPD patients were randomly divided into Tanreqing group, ambroxol hydrochloride group and control group. The patients in the three groups were all treated with conventional therapy. Furthermore, intravenous drip infusion of 20 ml Tanreqing injection (once daily) and 15 mg ambroxol hydrochloride injection (twice daily) were administered respectively to the patients in the Tanreqing group and ambroxol hydrochloride group. They were all treated for 10 days. Symptom score of traditional Chinese medicine (TCM), plasma concentrations of interleukin-8 (IL-8), IL-10 and neutrophil elastase (NE) were detected before and after treatment. RESULTS: Cough, sputum amount, expectoration, dyspnea, fever, coated tongue and pulse tracings were improved obviously in Tanreqing group (P0.05). Compared with ambroxol hydrochloride group and the control group, the coated tongue was improved obviously in Tanreqing group (P>0.05). After treatment, plasma concentrations of IL-8, IL-10 and NE were decreased in Tanreqing group and ambroxol hydrochloride group (P0.05). Total response rates in Tanreqing group and ambroxol hydrochloride group were higher than that in the control group (P0.05). There was no significant difference in total response rate among the three groups (P>0.05). CONCLUSION: Tanreqing injection can improve TCM signs and symptoms in AECOPD patients, and the mechanism maybe due to the decrease of serum levels of IL-8 and NE and improvement of IL-10 level.

ABSTRACT

OBJECTIVE: To investigate the mechanism of Chaiqin Chengqi Decoction (CQCQD), a compound of traditional Chinese herbal medicine, acting on the pancreatic acinar cell calcium overload in rats with acute pancreatitis (AP). METHODS: A total of 30 SD rats were randomly divided into normal control group, untreated group and CQCQD group (n=10, respectively). AP was induced in rats by caerulein (5x50 mug/kg) intraperitoneal injection within 4 h. The pancreatic tissue SERCA1 and SERCA2 mRNA expressions were detected by fluorescent quantization polymerase chain reaction method; intracellular calcium fluorescence intensity (FI) of pancreatic acinar cells and the pancreatic pathological score were measured by laser scanning confocal microscopy and light microscopy respectively. RESULTS: There were no SERCA1 mRNA expressions in pancreatic acinar cells of rats in the normal control group and the untreated group. The expression of pancreatic SERCA2 mRNA in the untreated group was down-regulated compared with that in the normal control group (expression ratio=0.536; P=0.001); the expression of pancreatic SERCA2 mRNA in the CQCQD group was up-regulated compared with that in the untreated group (expression ratio=2.00; P=0.012). The pancreatic pathological score in the CQCQD group was lower than that in the untreated group and the FI of Ca(2+) was also lower. CONCLUSION: CQCQD can up-regulate the expression of pancreatic SERCA2 mRNA, release the calcium overload, and hence reduce the pathological changes in pancreatic tissue.

ABSTRACT

OBJECTIVE: To evaluate the immuno-modulatory effect of short course administration of somatostatin (stilamin) continuously at early stage in patients with severe acute pancreatitis (SAP). METHODS: Thirty-nine patients with SAP (22 men, 17 women; the middle age was 49 years)were randomly allocated into control group (20 patients treated with non-surgical integrated traditional Chinese and Western medicine) and treatment group (19 patients treated with somatostatin administered intravenously at a dosage of 250 mug/h for consecutive 72 hours as well as the treatment for the control group). Laboratory parameters, including the expressions of CD(3), CD(4) and CD(8) in lymphocytes (tested by flow cytometry) and C reactive protein (CRP), and indexes of therapeutic effect, including the occurrence of organic dysfunction, local complication and mortality between the two groups were compared. Another group of 30 healthy volunteers (19 men, 11 women; the middle age was 47 years) were recruited for testing the normal levels of CD(3), CD(4) and CD(8). RESULTS: (1) The levels of CD(3), CD(4) and CD(4)/CD(8) in lymphocytes before treatment in both groups were significantly lower than those in the healthy subjects (P<0.05), but there were no statistical differences between the two groups. At the 4th day, CD(3), CD(4) and CD(4)/CD(8) increased significantly in the treatment group (P<0.05) while no changes in the control group; the levels of CD(4) and CD(4)/CD(8) in the treatment group were also higher than those in the control group (P<0.05). (2) The CRP levels of the 2 groups showed no statistical difference before and 4 days after the treatment, but the CRP level in the treatment group was significantly lower than that in the control group at the 7th day (P<0.05). WBC count, serum levels of amylase, lipase, lactate dehydrogenase, aspartate aminotransferase, as well as the score of APACHE II in the treatment group recovered more quickly than those in the control group (P<0.05). (3) The occurrences of organic dysfunctions, local complications and mortality in both groups were not statistically different. CONCLUSION: The short course administration of somatostatin continuously at early stage can reduce the inflammatory response, up-regulate the cell immune function and improve the conditions of the patients with SAP, but its effect on mortality and morbidity needs further study.

ABSTRACT

This investigation was amied at the infiltrative capability of the machinable-infiltrated-ceramic(MIC) glass and the color of the composite after the MIC glass infiltrated through the aluminous matrix with different packing densities. By heating the components to 1100 degrees C for 2 hours, the MIC glass was made to infiltrate through the aluminous matrix with different packing densities. We measured the infiltrative depth and the color parameter and observed the rupture surface of the composite by means of SEM. There was a linear relation between the square of infiltrative depth and the packing density of aluminous matrix. The minimal depth was 3.092 mm. No relationship was noted between the composite's color coefficient and the packing density of aluminous matrix. In the rupture process of the composite, crack deflexion, crystal evulsion, and rupture through crystal could be observed. This experiment proved that the infiltrative characters of MIC glass meet the clinical requirement, the composite's color is steady and the mechanical intensity is stable.

ABSTRACT

In order to develop tinted infiltration glass and its colorants, which can make the ceramic have good spectrum transmittance, color space, color stability and be suitable for clinical use, we selected the best prescription and confirm the ingredient and content of the colorants. Molten glass was prepared in Al2O3 crucible by heating the components to 1420 degrees C for 2 hours. The refractive index and thermal stability of the glass were investigated. The refractive index of the MIC infiltration glass was 1.5969(587.6 nm, nd). It was close to the index of aluminous matrix 1.759(546.07 nm, ne), which increased the spectrum transmittance of MIC. The thermal expansion coefficient of the glass was 7.565 microns/m/degree C, which was compatible with the thermal expansion coefficient of aluminous matrix(8.214 microns/m/degree C). This study proved that the tinted infiltration glass has good color stability, spectrum transmittance, and thermal expansion properties.

ABSTRACT

The pharmacological characteristics of recipes are multi-components, multi-ways, multi-links, and multi-targets, on which the efficacy substances in recipes are worth of studying. To explore the essential substances of Chinese herbs in recipe is one of the key points for modernization of Chinese materia medica. The pharmacological effects and bioactivities inside the target tissue of lesion body are still not clear, even though there are some progress in herb serum pharmacology, recipe pharmacokinetics, and pharmacodynamics. The main effective components of recipe to pancreas in acute pancreatitis models by ig or enema were investigated by HPLC-MS method so as to establish a new hypothesis (tissue pharmacology of recipe): i.e. guided hy fundamental theory of traditional Chinese medicine to study the effect and the mechanism of recipe's essential substances on target tissues with pharmacological methods; to establish the experimental method for searching the foundation, the principle, and the direct substances of recipes which act on target tissues; to form more effective formula if possible, and to explore in a new researching field for the recipes of Chinese materia medica.
